To drag this kicking and screaming back on topic and answer OP's original question:
If being in the U.S. is a requirement, I'd pick SF, Seattle, or NYC, provided I could stand the cost of living and the rest of the lifestyle that goes with living in such a place. (Not owning a car, small apartment, possibly living with roommates, etc.) If you need a cheaper cost of living, Austin might be an option, or, for that matter, any college town in the South.
Colorado has a ballot initiative being voted on tomorrow that, if it passes, would pretty much legalize recreational pot in that state, if I'm understanding things correctly. Might make Denver or Boulder a good option.
If you can live outside the U.S. and get permission to live and work in another country? Most of the big cities of Europe are fairly socially tolerant. Amsterdam, Zürich, Berlin, Madrid, provided you can pick up enough of the local language. Maybe Montréal, if access to the U.S. is a concern. You'll have the same cost-of-living concerns in all those places that you will in NYC or SF, though.

well from the standpoint of legalized pot...Washington state or Oregon is the place to be. they have the largest allowance of legal pot by 3x over any state (except for maybe Connecticut which would allow a 1month supply but that's anyone's guess what that means.

Having lived there for 20 years I vouch for the San Francisco Bay Area. It is usually rated as the most LGBT-friendly area in the entire US. I would also put in for any of the major cities on the "Left Coast": SF, Portland, Seattle, Vancouver (BC) though these are all very expensive to live in (Vancouver has been rated the second-most expensive city to live in--in the entire world). If you're into cross-dressing publicly, then SF definitely has it over anywhere else. All the major street fairs are LGBT-run, too. And you're only 6-8 hours drive away from Los Angeles.

As for pot-friendly, CA may not be as progressive yet as some other states, but its only a matter of time, and probably not much at that. I'm not a smoker but to my knowledge most municipalities around the Bay Area, and other major cities, practically ignore pot laws for individual smokers (trafficking is another matter). There are exceptions, of course, but some research would tell you where the enforcement is most lax.

hey...SF will have nothing on NYC come next year when we elect our first lesbian mayor...NYC has the largest population (by gross number, not percentage) of LGBT identifying people.
